제목 | Chitosan nanoparticles based on polyelectrolyte complexes of all-trans retinoic acid and free-amine water-soluble chitosan |
초록 | Chitosan is a biodegradable, biocompatible, naturally occurring polymers consisting of 2-amino-2-deoxy- -D-glucan. The chitosan has both reactive amino and hydroxyl groups that can be used to chemically alter its properties under mild reaction conditions. There have been many chitosan derivatives and polyelectrolyte complexes with anionic agents for biomedical application.All-trans retinoic acid (atRA) is effective in the treatment of epithelial and hematological malignancies such as breast cancer, head and neck cancer, ovarian adenocarcinoma, and acute promyelocytic leukemia (APL). However, some side effects such as retinoid acute resistance, hypertriglyceridermia, mucocutaneous dryness, and headache were reported in spite of its pronounced effects limits the clinical applications of atRA. Moreover, poor aqueous solubility of atRA are also problematic for clinical applications. In this study, we synthesized water soluble chitosan with low molecular weight and free amine group(LWWSC). By using this, polyelectrolyte complexes between atRA and LMWSC for application of novel antitumor drug delivery system. Polyelectrolyte complexes (PEC) nanoparticles of atRA and LMWSC prepared by dialysis method was shown in nice spherical shape and their particle size were varied to 50-500 nm according to preparation variables. AtRA was continuously released from PEC over one month. |
