Freestanding lipid bilayer with large area and planar geomtry provides an outstanding in-vitro model platform for a number of biological membranes. However, it has been extremely difficult to achieve the lipid bilayers to date due to its poor stability. We present a novel technique to form a stable, freestanding, solvent-free lipid bilayers with large area and planar geometry. Our new technique is based on the hybridization of a droplet monolayer and a planar monolayer. With the help of a water in oil emulsion stabilizer, we were able to create milimeter-scale stable freestanding lipid bilayers. We also discovered an origin of this poor stability and set up stability criteria based on two theories, the entropy of mixing and the spontaneous curvature of lipids and surfactants. Finally, several different applications using our newly developed lipid bilayers will be discussed. |
