Single process O/W/O emulsion was invented to prepare Paclitaxel(PTX)-loaded gelatin nanoparticles. Linoleic acid was chosen among natural fatty acids as exterior medium for single emulsion process. The size-mean and zeta potential of PTX-loaded gelatin NPs in their suspension were 164.95 nm (±6.43 nm) distributed with a polydispersity of 0.074 (±0.046) and -23.85 mV (±12.66 mV), respectively. The size of PTX-loaded gelatin NPs prepared in this study was the smallest among the reported sizes PTX-loaded gelatin NPs, which would contribute to the effect of enhanced permeability and retention (EPR). The slowly sustained PTX release of day-time scale without an initial burst release into a release medium was observed to accompany a delay of more than 2 days before bursting PTX-release from 50 hr to 70 hr in spite of adding protein degrading enzyme. PTX loading efficiency was observed to be 81%. This loading efficiency was greater than those of any previous investigation on PTX-loaded gelatin NPs. |
