제목 | Development of Three-arm junction RNA nanostructures as Dicer substrate for enhanced in vivo gene silencing |
초록 | Three-arm junction RNA nanostructures(Y-RNA) have enhanced in vitro and in vivo RNAi potency compared to canonicalsiRNA. Y-RNA was structurally optimized to be Dicer substrate and load into RISCin the proper orientation. In addition, FP assay was used to determine bindingaffinity with RLC components such as TRBP, Dicer, and Ago2. TRBP is an integralcofactor of Dicer, which distinguishes Dicer substrate RNA for effectiveloading and transfer to Ago2. We found that Dicer substrate RNA binds to TRBPwith a 35-fold higher affinity than siRNA. This allows that Y-RNA to formactive RISC at much lower concentrations. Further, we have developed auniversal chemical modification for Y-RNA to be applicable clinically. Theposition-specific chemical modification of Y-RNA did not interfere with bindingto the RLC components. In addition, it offered the enhancement of serumstability and in vivo RNAi potency. |
