제목 Glucose-capturing polymer nanoparticles for cancer theranosis
초록 Since conventional cancer therapeutics may induce side effects such as renal disorder, hair loss, etc., functionalized polymer nanoparticles have been extensively investigated to provide new opportunity for targeted delivery of bioactive agents. Most tumor cells show high aerobic glycolysis due to critical damage of mitochondrial respiration and hypoxia stress in the cells. We thus hypothesized that glucose-capturing nanoparticles could be effective to induce glucose deprivation of cancer cells, leading to apoptotic cell death. Phenylboronic acid, which is known to reversibly bind to diols of glucose, was chemically introduced to the polysaccharide backbone and nanoparticles were prepared via self-assembly in water. Various physiochemical properties, anti-glycolysis effect, and apoptosis induction of glucose-capturing nanoparticles were investigated in vitro. We also tested the efficacy of the nanoparticles as a theranostic system using a tumor-bearing mouse model.
