Low molecular weight bPEI is generally accepted as one of efficient polymeric carriers with low toxicity. Guanidinium group has been known to improve transfection efficiency due to possible hydrogen bonding with DNA phosphate anions, resulting in higher cell membrane-penetrating properties. Moreover, folate binding allows to promote target-specific delivery in folate-receptor over-expression cells. In this study, bPEI with a guanidinium group was successfully synthesized by chemical modification under favorable conditions, coupled to folate via physical interaction of adamatane-cyclodextrin bridge to exhibit higher cell penetrating efficiency simultaneously with specific-targeting. The formation and degree of substitution of modified bPEI were confirmed by NMR, ESI-MS and EA. The physicochemical properties of modified bPEI were evaluated by particle size measurement and z-potential assay. Also, promoted uptake of modified bPEI was investigated in variety of folate-receptor tumor cells. |
