Improving therapeutic angiogenesis in human adipose derived stem cells with blue light for treating hindlimb ischemia |
초록 |
Low level light therapy (LLLT) has drawn attention as a new therapeutic method for treating various disease. We introduced blue organic light emitting diode (bOLED) as a safe and effective light source based on low heat generation and luminance compared to the conventional light source. Here in, we optimized the luminance, wavelength and light fluence of bOLED and showed molecular mechanisms which enhance therapeutic angiogenic efficacy of human adipose derived stem cells (hADSCs) by controlling ROS generation of hADSCs. With the optimized conditions of bOLED light, biomodulation of hADSCs was increased without phototoxicity. It was also discovered that angiogenic expression of hADSCs was promoted attributed to mild ROS generation when irradiated with bOLED light. Tissue regeneration was promoted when bOLED light irradiated hADSCs were transplanted to the hindlimb ischemia mouse model. These findings might open new method of blue light in LLLT for ischemic disease. |
