제목 | Self-assembled DNA/Polymer Hybrid Micelles for Gene Therapy |
초록 | Polymeric micelles have been considered as a promising delivery system for drugs as well as nucleic acids. In our previous study, the oligonucleotide-PEG (ODN-PEG) conjugate could form polyelectrolyte complex micelles by interacting with various cationic polymers, such as KALA peptide, poly(L-lysine), PEI, and protamine. The conjugate of antisense c-raf oligonucleotide (ODN) and poly(ethyleneglycol) (PEG) was synthesized for intracellular ODN delivery in the form of micelles. The ODN-PEG conjugates could associate to form polyelectrolyte complex micelles when complexed with polyethylenimine (PEI) in an aqueous solution. Effective hydrodynamic volume of the micelle was ca. 80 nm with narrow size distribution. The intracellular transport efficiency of the micelles was much higher than that of unmodified ODN itself. The polyelectrolyte complex micelles fabricated from a physical mixture of antisense c-raf ODN-PEG conjugate and PEI showed superior anti-proliferative activity against ovarian cancer cells in vitro as well as in vivo. |
