Core-Functionalized Pluronic Micelles: Doxorubicin Conjugation in Micelle Core via Acid Cleavable Linkage |

Novel polymeric micelles, self-assembled from a biocompatible triblock copolymer of poly(ethylene oxide) and poly(propylene oxide) (PEO-PPO-PEO) having multiple hydroxyl groups in the middle segment, were prepared to solubilize moderately soluble drug doxorubicin (DOX) by conjugating the drug via acid cleavable linkage. The functionalized triblock copolymer was synthesized using secondary amine mediated epoxide ring-opening reaction. The resultant polymer self-assembled into micelle in aqueous solution having a diameter of 88.5±6.3 nm and showed extremely low toxicity. DOX molecules were conjugated to the copolymer via hydrazone linkage to produce DOX conjugated copolymer. Release of DOX was pH-dependent showing higher release rate at pH 5.0 compared with that at pH 7.4, suggesting that the micelles specifically release DOX in endosomal environment. They also exhibited higher cytotoxicity than free DOX indicating that they were more capable of cell uptake than free DOX. |
