제목 Development of Pheophorbide A-conjugated Nanoparticles for Photodynamic Therapy
초록 An effective tumor-targeted drug delivery system for photodynamic therapy was developed by designing ligand-mediated nanoparticles with stable formulations of a hydrophobic photosensitizer. A high-performance photosensitizer Pheophorbide a (Pheo) was conjugated to folic acid-conjugated biocompatible block copolymer of polyethylene glycol (PEG) and poly aspartic acid (P(Asp)) through pH-sensitive hydrazone linkage. Photosensitizer-loaded amphiphilic copolymeric nanoparticles (FA-PEG-P(Asp-Hyd)-Pheo) were prepared by micelle formation in aqueous solution. The solubility and photophysical properties of hydrophobic Pheo been significantly improved. The in vitro release profiles of Pheo from the nanoparticles were strongly influenced by pH and the amount of photosensitizer that was loaded. In vitro tests using HeLa cells indicated that the FA-PEG-P(Asp-Hyd)-Pheo nanoparticles had excellent cellular localization and killed HeLa cells effectively compare to free Pheo under laser radiation.
