Monoolein (MO) in water takes on bicontinuous inverted cubic phase, depending on the temperature and the concentration. Since the cubic phase of MO is composed of lipid matrix and water channel, both hydrophobic and hydrophilic active ingredients could be accommodated in the self-assembly. Cubic phases are non-toxic, biodegradable, biocompatible, adhesive to skin and mucus. in this study, a hydrophilic drug and poly(N-isopropylacrylamide)(PNIPAM) was included in water channel to develop a novel temperature and pH-sensitive delivery system. A cubic phase was prepared by hydrating MO using an aqueous solution of PNIAPM copolymers. As a copolymer P(N-isopropylacrylamide-co-octadecyl acrylate)(copolymerⅠ) and P(N-isopropylacrylamide-co-octadecyl acrylate-co-methacrylic acid)(copolymerⅡ) were employed. The characteristics of the copolymers are that copolymer I is temperature-sensitive, and copolymer II is both pH- and temperature-sensitive. |
