제목 Preparation of Gemcitabine-Loaded Poly(L-lactide) Microparticles Using Supercritical Fluid: Effect of Additives
초록 Gemcitabine is an effective antitumor agent in the treatment of several solid tumors. However, it has been reported that gemcitabine has severe problems such as short plasma half-life and rapid metabolism. In this study, microparticles of poly(L-lactide) (L-PLA) loaded with gemcitabine were prepared by a supercritical fluid process called aerosol solvent extraction system (ASES) utilizing supercritical CO2 as antisolvent. To achieve an improvement in drug release profiles, varieties of gemcitabine-loaded L-PLA microparticles containing such additives as 2-hydroxypropyl-β-cyclodextrin, ascorbic acid, L- and DL-lactide were prepared using the ASES technique. The morphology, entrapment efficiency, drug loading, and in vitro drug release of these microparticles were examined to evaluate the influence of various additives.
