Gemcitabine is an effective antitumor agent in the treatment of several solid tumors. However, it has been reported that gemcitabine has severe problems such as short plasma half-life and rapid metabolism. In this study, microparticles of poly(L-lactide) (L-PLA) loaded with gemcitabine were prepared by a supercritical fluid process called aerosol solvent extraction system (ASES) utilizing supercritical CO2 as antisolvent. To achieve an improvement in drug release profiles, varieties of gemcitabine-loaded L-PLA microparticles containing such additives as 2-hydroxypropyl-β-cyclodextrin, ascorbic acid, L- and DL-lactide were prepared using the ASES technique. The morphology, entrapment efficiency, drug loading, and in vitro drug release of these microparticles were examined to evaluate the influence of various additives. |
