Messenger RNA (mRNA) is a genetic material generated by transcription of DNA and these single strand RNA (ssRNA) molecules are employed to guide the target protein production in the cell. mRNA can be an alternative genetic materials to regulate cellular behavior or phenotype instead of large plasmid DNA (pDNA). It has multiple advantages over pDNA such as 1) fast on-set of target gene expression, 2) less genetic stress on host cells, 3) no requirement for nuclear localization. In this study, we have prepared mRNA by in vitro transcription (IVT) methods along with polymerase chain reaction (PCR) and circularization template DNA. Various bioreducible polymeric systems have been screened to optimize to their mRNA delivery efficiency. Transfection and expression of target genes have been confirmed in mouse embryonic fibroblast (MEF) cells. |
