제목 Ocular delivery of brimonidine tartrate using thermo-gelling polymer for glaucoma therapy
초록 Conventional eye drops are often eliminated rapidly after administration, leading to a short residence time in the pre-corneal area and a limited transcorneal absorption. Here, a new thermo-sensitive hexanoyl glycol chitosan (HGC) was synthesized and utilized to develop a HGC based eye drop formulation. The thermo-sensitive sol-gel transition of HGC was controlled in a temperature range of 25 to 50 ℃ by modulating the degree of N-hexanoylation. The MTT assay and Live/Dead assay showed that HGC has low cyto-toxicity at the concentration of 0.25-1.25 wt%. The in vivo ocular delivery efficiency of brimonidine tartrate using aqueous HGC solution (BRT-HGC) were evaluated and compared to that of Alphagan P using rabbit. The intraocular pressure lowering effect of BRT-HGC solution was extended to 14 hr. The HGC formulation demonstrated enhanced preocular retention due to thermos-sensitive viscosity increase, improving the drug absoption.
