Hydrogels are of great interest as potential biomaterials for sustained delivery of various therapeutic agents. In this study, we designed a novel blend system hydrogel with high mechanical properties for islet cell delivery. Firstly, Pluronic F127, PEO-PPO-PEO triblock copolymer, was simply modified into two types of functional groups, thiol and pyridyl disulfide, respectively. In physiological environment, gelation occurs firstly via micelle packing mechanism of Pluronic F127 and thiol-disulfide exchange reaction which can be thought as a driving force of mechanically stable hydrogel system happens at the shell of micelles. The properties of resultant chemically as well as physically crosslinked Pluronic hydrogels were confirmed by sol-gel phase diagram, in vitro and in vivo experiment. Further, by in vivo experiment, it is expected that this hydrogel system can be applied to islet cell delivery system for controlling high glucose level in blood. |
