This work reports a highly sensitive cTnI biosensing platform for point-of-care testing, named PASCAL (Programmed Amplification using Swellable Polymer for Chemiluminescence-Assisted Lateral Flow Immunoassay). The PASCAL reveals a pioneering analytical sensitivity over wide dynamic range by incorporating gold nanoparticles in a polymeric network of peroxidase. Moreover, an integration of a mass-producible functional module based on a water-swellable polymer into the PASCAL allows for automating immunoassay and signal-amplifying reactions in a single device with programmed intervals and without the need for an external power source. The PASCAL enables high performance testing of human cTnI standard/clinical samples, with high-sensitivity (0.39 pg mL-1) and precision (coefficient of variation <10% at the 99th percentile cTnI level) and within acceptable clinical guideline, and with an assay time of 20 min. |
