제목 Photocrosslinkable 3D microenvironments functionalized with bioactive molecules for cartilage tissue engineering
초록 In-situ gelling systems can be formed into any desired shape of defect at the site of injection. These hydrogels allow for the delivery of cells and bioactive molecules to the injured tissue. Photocrosslinkable and biodegradable hydrogels have been widely used in biomedical applications. Both bioactive molecules and physical signals in ECM play critical regulatory roles in differentiation of MSCs. In this study, we functionalized photocrosslinkable hydrogels with TGF-β or Wnt signaling molecules. The crosslinking density controlled by macromer/initiator concentration or light exposure time could modulate mechanical properties of photocrosslinkable hydrogels. We evaluated their role in regulating chondrogenesis by encapsulating ASCs. Cells maintained great viability in 3D hydrogels during in vitro culture. In addition, the biological 3D microenvironment with tailorable mechanical properties, regulated chondrogenic differentiation of encapsulated ASCs via TGF-β or Wnt signaling.
