Cellular imaging is used for successful cell therapy because the fate of the injected cell can be tracked and visualized. Among various imaging tools, magnetic resonance imaging (MRI) is favorably used due to its higher resolution through MR contrast agents. On the other hand, the injected cells can induce the nonspecific inflammation due to the activation of platelets in the bloodstream. Here, we developed the bi-functional MR contrast agent for cell imaging and for inhibiting the inflammation. To this end, we newly developed heparinized SPIO nanoparticles with higher magnetic moment. For cell labeling, the heparinized SPIO nanoparticles were chemically conjugated onto cell surface. This chemical modification using heparinized SPIO nanoparticles did not affect the viability and function of the cells. Also, the modified cells could be well visualized using MRI in vitro and in vivo. In addition, the heparinized SPIO nanoparticles could inhibit the activation of platelets, thereby attenuating the inflammation in the bloodstream. Therefore, it could enhance the viability and function of the cells in the body. Collectively, we developed highly efficient heparinized SPIO nanoparticles. |
