Low molecular weight bPEI conjugated superparamagnetic nanoparticle (NP) for efficient siRNA delivery |

Magnetofection is a novel method based on the use of magnetic NP. It allows to target cells and triggers nucleic acids uptake. Superparamagnetic particles are strong magnetization and it shows little cytotoxicity. Also it reduces the time to achieve efficient transfection. But it requires another mediator which can condense siRNA and protect it from degradation. PEI is a class of cationic polymers proven to be effective for gene delivery. In this study, low molecular weight bPEI(1.8kDa) was covalently conjugated with magnetic NP(superparamagnetic iron oxide,SPION) by EDC/NHS chemistry. The physico-chemical properties of bPEI1800-SPION were evaluated by various methods. The bPEI1800-SPION showed complete siRNA condensation. Inhibition of VEGF expression was confirmed in the presence of a magnetic field for even a short period of time. Therefore bPEI1800-SPION is a valuable siRNA transfer vehicle that can lead very rapid and efficient gene transfer in vitro in a magnetic field. |
