제목 Hybrid Polymer-Superparamagnetic Iron Oxide Nanoparticles Mediated Highly Efficient Magnetofection
초록 Magnetofection is a mgnet-mediated transfection method using magnetic nanoparticles(MNP). This method reduces the time to achieve enhanced transfection. For efficient gene expression, however, it requires another mediator which condense gene and induce endosomal escape. PEI is one of the most effective cationic polymer for gene delivery. In this study, the efficacy and mechanism of polymer-MNP as gene delivery vehicles were evaluated by magnetofection. The BPEI-SPION was synthesized by conjugation of LMW BPEI with superparamagnetic iron oxide nanoparticle(SPION). For short incubation time or in serum-containing media, the BPEI-SPION/pDNA magnetoplex was able to highly transfectgene into cells in a magnetic field. Also in a magnetic field, the magnetoplex was accumulated rapidly at the cells but no effect on the cellular uptake mechanism. Thus, the BPEI-SPION is intelligent gene delivery vector which exhibits anti-fouling properties and fast, efficient transfection in a magnetic field.
