The simultaneous extraction and separation of matrine, sophoridine and sophocarpine from sophora flavescens Ait was developed by liquid-liquid extraction with liquid chromatography separation. The optimum extraction conditions were established using different extraction solvents, procedures and times. The extracts from sophora flavescens Ait were separated and determined using a C18 column with methanol/water/diethylamine (55/45/0.10, v/v/v) as the mobile phase. Different preparative columns with packing sizes ranged from 12 to 63 μm were used to isolate the three compounds from the extracts, and the column with a 12 μm particle size showed better separation. The amounts and the recoveries of sophocarpine, sophoridine and martrine obtained from sophora flavescens Ait were 0.072 mg/g, 0.035 mg/g and 0.037mg/g, and 83.42%, 85.11% and 74.20%, respectively. |
