Many bisindole molecules isolated from natural sources have potent antitumor and antiviral properties. One of the best known bisindole molecules, violacein is also naturally occurring a purple pigment and is widely used in the cosmetic and pharmaceutical fields. In this study, we constructed violacein biosynthetic pathway in Escherichia coli by monocistronically expressing each gene of the violacein-synthesizing operon vioABCDE originated from Chromobacterium violaceum. To increase the productivity, we first engineered 5’-UTR region of each gene to improve the expression of genes in the biosynthetic pathway. We then explored the optimal culture conditions and medium composition for improved violacein production. In addition, we engineered host metabolism to reroute the metabolic fluxes to the precursor molecule, tryptophan. This rational metabolic engineering approach can generally be applied to produce various bisindole molecules. |
