ROS-responsive drug delivery systems have limitation in that the drug release uncontrollable, which could lead to side effects and limit therapeutic efficiency owing to premature drug release in the blood circulation and slow diffusional release at the tumor site. To overcome these limitations, we prepared photo-mediated ROS-generative polymeric micelles (PGM). ROS-responsive polymer and photosensitizer (PS) were conjugated. When irradiated with specific wavelengths, PS generates ROS, polymer’s property can change in ROS condition. And then the encapsulated anticancer drug could be released. And ROS-induced photochemical membrane disruption allowed for cytosolic release from endo/lysosomes. This PGMs showed significantly enhanced cytotoxicity in cancer cells compared to PGMs without light exposure. In tumor bearing mouse, PGM treated group’s tumor sizes were significantly decreased. |
