Herein, we first report the feasibility of stable and consecutive targetable nanocontainers carrying molecular beacons for the intracellular recognition of miR-34a expression level in human metastatic breast cancer cells. In particular, hyaluronic acid (HA) is non-toxic, non-immunogenic, and has high affinity against adhesion molecule CD44, a critical marker of cancer stem cells in breast cancer. Thus, our sequential targeting system which targets specific miRNA by molecular beacon via specific CD44 route-mediated endocytosis enables intracellular imaging. Moreover, targeted detection ability by optical imaging and enhanced loading efficiency as well as sustained release of miR-34a beacons were achieved using HA-coated nanocontainers (HNCs) containing miR-34a beacons (bHNCs). |
