Biomacromolecular chaperone Refolding of denatured proteins mediated by the self-assembled nanoparticles |

Protein refolding is the crucial process by which a misfolded protein assisted by molecular chaperones reconfigures to its characteristic functional active state. Here we have synthesized hydrophobically modified hyaluronic acid (HA) by chemically conjugating cholanic acid to HA backbone, in which the conjugate could form nanoparticles by self-assembly of cholanic acid. The physiochemical characteristics were investigated by 1H NMR, dynamic light scattering, flourescence spectrophotometer. The chaperon-like activity was evaluated using carbonic anhydrase B (CAB) as a model protein and β-cyclodextrins as a releasing agent. This artificial chaperone can be useful as protein stabilizing and sustained delivery system. |
