Development of mannan-decorated thiolated Eudragit microspheres for an efficient nasal vaccine delivery |

We decorated thioated Eudragit microspheres with mannan for targeting mannose receptors (MR) in antigen presenting cells (APCs) to obtain the systemic and mucosal immune responses through nasally administrated ovalbumin (OVA) as a model vaccine. Mannan-decorated thiolated Eudragit microspheres (Man-TEM) and thiolated Eudragit microspheres (TEM) were prepared using a double emulsion method (W/O/W) and characterized them physicochemically. The APC cell lines were used to validate and clarify the uptaking efficiency and mechanism by CLSM and FACS analysis. As a result, Man-TEM had higher uptaking ability than TEM, indication of MR receptor-mediated endocytosis mechanism. Nasal vaccination of OVA-loaded Man-TEM in mouse showed higher levels of serum IgG and mucosal IgA than the TEM-treated group due to the tageting ability of mannose in the Man-TEM. These results suggest that Man-TEM may be a promising candidate for a nasal vaccine delivery system to elicit systemic and mucosal immunity. |
