제목 Development of mannan-decorated thiolated Eudragit microspheres for an efficient nasal vaccine delivery
초록 We decorated thioated Eudragit microspheres with mannan for targeting mannose receptors (MR) in antigen presenting cells (APCs) to obtain the systemic and mucosal immune responses through nasally administrated ovalbumin (OVA) as a model vaccine. Mannan-decorated thiolated Eudragit microspheres (Man-TEM) and thiolated Eudragit microspheres (TEM) were prepared using a double emulsion method (W/O/W) and characterized them physicochemically. The APC cell lines were used to validate and clarify the uptaking efficiency and mechanism by CLSM and FACS analysis. As a result, Man-TEM had higher uptaking ability than TEM, indication of MR receptor-mediated endocytosis mechanism. Nasal vaccination of OVA-loaded Man-TEM in mouse showed higher levels of serum IgG and mucosal IgA than the TEM-treated group due to the tageting ability of mannose in the Man-TEM. These results suggest that Man-TEM may be a promising candidate for a nasal vaccine delivery system to elicit systemic and mucosal immunity.
