제목 In vivo and in vitro study of PLA-based scaffolds for rat muscle derived stem cells
초록 The use of three-dimensional scaffolds to mimic biological functions like the extracellular matrix is important in the field of tissue engineering. This study makes a comparative study on PLA microfiber prepared by electrospining and PLA scaffold prepared by salt-leaching. Comparison of PLA microfiber and PLA scaffold was carried out with SEM, porosimeter and contact angle. Cellular viability was assayed to examine attaching and proliferation of rat muscle derived stem cells on the PLA microfiber and PLA scaffold. This assay resulted the PLA microfiber have less toxicity than the PLA scaffold. Then PLA microfiber and PLA scaffold were implanted subcutaneously into Fischer rats and observed the effect for 4 weeks. H&E staining showed a formation of new blood vessels in only PLA microfiber. ED1 staining showed that PLA microfiber have less macrophages than PLA scaffold. In conclusion, this study shows that PLA microfiber is more biocompatible than PLA scaffold in vivo.
