제목 Neuronal Differentiation of Rat Muscle-Derived Stem Cells in Chitosan-Based Hydrogels
초록 Muscle-derived stem cells (MDSCs) can be isolated from skeletal muscles and MDSCs have characteristics of self-renewal capacity, proliferative potential, and differentiation. We chose valproic acid (VA) for inducing neural differentiation of MDSCs in chitosan-based hydrogels. Chitosan gel surfaces were prepared by loading the liquid solution into clean well plates and maintaining the temperature at 37oC for 24 h. Each gel was then seeded with rMDSCs. After pre-induction with basic fibroblast growth factor and subsequent treatment with VA. The rMDSC on chitosan hydrogel in the presence of VA showed higher expression for neuron-specific enolase (NSE) and beta tubulin III (Tuj-1) as neural markers, oligodendrocyte transcription factor 2 (Olig-2) as an oligodendrocyte marker, and glial fibrillary acidic protein (GFAP) as an astrocyte marker. Our results suggest that rMDSCs on chitosan hydrogel in the presence of VA can be differentiated into a neural-like phenotype.
