Hydrogel-based signal amplification technologies for in vitro diagnostics and prognosis |

Detection of tiny amounts of potential biomarkers from various body fluids such as urine and blood has continuously drawn attention. We present two specific examples of detection of biomarkers without modifying targets for in vitro diagnosis and prognosis. We introduce hydrogel-based hybridization chain reaction (HCR) for multiplex signal amplification to detect urinary exosomal miRNAs from human clinical samples. We identified small amounts (~amol) of exosomal miRNAs from 600 μL of urine with up to ~35-fold amplification. Furthermore, we proposed ratiometric analysis without requiring normalization to a reference miRNA. We also introduce Sequence-specific nanoparticle With Interpretative Toehold-mediated sequence deCoding in Hydrogel (SWITCH) for multiplex SNP genotyping. The encoding with gold nanoparticle probes transduces each SNP target to ~1000 invaders with prominently different sequences between wild and mutant types, featuring polymerase chain reaction (PCR)-free amplification. We demonstrated 4-plex detection of the warfarin-associated SNP targets spiked in commercially validated human serum with excellent specificity. |
