Development of rapid diagnosis of active Tuberculosis using bio-nano sensor technology |

Mycobacterium tuberculosis (MTB) is a serious infectious pathogen causing tuberculosis (TB). The TB has become a global threat leading people to deaths especially in developing countries. To prevent TB spreading and also improve patient care, a rapid and accurate diagnosis method of TB is required for its early diagnosis. In this study, one of the secretory antigens of MTB was chosen as a target for early TB diagnosis. Several recombinant antibody fragments against the targets were expressed in Escherichia coli. These antibodies were analyzed for their binding affinities with the target through surface plasmonic resonance analysis and enzyme-linked immunosorbent assay. The binding properties of the gold-conjugated antibody with the targets in urine samples, proving its great potential as an early-stage diagnosis for active TB, were confirmed through sequential signal enhancement of colorimetric analysis and proved its efficiency as a detection tool with naked-eye. |
