제목 Synthesis of DNA-directed synthesized gold nanoparticle for application of DNA point mutation detection
초록 Distortions in the binding affinity of site-specific protein to DNA are associated with genetic diseases, but high-fidelity methods for establishing a consensus rank of protein affinity for mutant DNA—particularly single-point mutations—for the purpose of early diagnosis are lacking. Here we report non-labeling endpoint detection and dynamic analysis of the binding of the mismatch repair initiation protein MutS to the eight most common single-point mutations in BRCA1 using a single-nanoparticle microfluidic platform comprising gold-bridged nanoparticles produced by direction-specific synthesis that can directly transduce information on the MutS-DNA interaction. Mutant DNA was classified into four types based on MutS binding affinity for various point mutations. This powerful method can be used to profile mutant DNA, analyze protein-DNA interactions, and elucidate the mechanisms of gene regulation in disease states.
