Preparation of Chitosan Microsphere Using Pluronic as a Stabilizer for a Nasal Vaccine Delivery System |

Chitosan is a valuable excipient for nasal drug delivery system due to its biocompatibility, biodegradability and low cost. Also, it has been reported that chitosan could induce cytotoxic T lymphocytes and macrophage activation. In this study, chitosan microspheres using pluronic as a stabilizer(PCMs) were prepared based on ionic gelation process of tripolyphosphate to increase stability of microsphere and to enhance immune activity and then, the PCMs loaded with Bordetella Bronchiseptica Dermonecrotoxin (BBD), a virulence factor of a causative agent of atrophic rhinitis, were prepared for nasal vaccination. The particle sizes of BBD-loaded PCMs were smaller than 10 μm and had narrow size distribution with uniformity. Mechanical stability of chitosan microsphere (CM) was enhanced using pluronic. BBD from PCMs was faster released than CMs. Also, more and longer tumor necrosis factor-α (TNFα) and nitric oxide (NO) from BBD-loaded PCMs were secreted than BBD itself. Furthermore, PCM/BBD stimulated more macrophage and showed higher response than BBD itself. |
