Recently, brown seaweed containing alginate as the major carbohydrate has been considered as renewable biomass for the production of biofuels and biochemical. In this study, we investigated alginate saccharification using the recombinant oligoalginate lyase (OAL) of Sphingomonas sp. MJ-3 to obtain alginate monosaccharide, unsaturated uronate. Saccharification efficiency greater than 70 % was obtained by batch saccharification of 1 %(w/v) alginate using 100 μg/ml of the recombinant Escherichia coli cell lysates containing MJ-3 OAL at pH 6.5 and 40 °C. Under fed-batch and batch mode, the saccharification efficiency was obtained 52.8 % and 68.5 %, respectively, from 4 %(w/v) alginate substrate using 400 μg/ml of the recombinant E. coli cell lysates at pH 6.5 and 40 °C. |
