제목 Oral hepatic cancer therapy by gold nanoparticle based siRNA complex with bile acid mediated glycol chitosan using enterohepatic circulation
초록 In this research is focus on oral hepatic cancer therapy by AuNP based siRNA complex coated by Taurocholic acid (TCA) mediated glycol chitosan(GC). TCA, one kind of bile acid is usually used for oral drug delivery system that could reach to liver by enterohepatic circulation. TCA mediated GC confirms by H-NMR and zeta-potential. Gel electro-phosphate and TEM analysis indicated AuNP based siRNA complex was made successfully and that complex stability and release property in the buffer solution. Caco-2 cell and HepG2 cell transport and uptake study indicated our carrier can pass to Caco-2 monolayer and uptake on the HepG2 cells. in vivo studies of fluorescence image display our carrier uptake in the small intestine and successfully accumulated in the hepatic cancer cells. Therefore our AuNP based siRNA complex coated TCA mediated GC can pass by epithelial cell in the small intestine and accumulation on hepatic cancer cell for therapy.
