제목 Poly(3-hydroxybutyrate) Production by High Cell Density Culture of Ralstonia eutropha: Oxygen Supply and Consumption
초록 A comparative fed-batch culture study of poly(3-hydroxybutyrate) (PHB) production by Ralstonia eutropha (R. eutropha) was carried out with air and high purity oxygen in 5-L, 30L, and 300-L fermentors. In the 30-L fermentor, the final dry cell weight and PHB concentrations obtained with high purity oxygen were 185.9 and 138.7g/L, which increased 3.8 and 6.2 times respectively as compared with the results obtained in the case of air supply. Whereas, the dry cell weight and PHB concentration in the 5-L fermentor increased 2.2 and 2.9 times respectively. They were mainly attributed to the enough oxygen supply in the case of high purity oxygen. It is impossible to meet the oxygen demand of cells in a large-scale high cell density culture by supplying air only. It was found that there was just about 4~5% oxygen consumed in the case of air supply, but it increased to 9~11% in the case of high purity oxygen supply.
