An α,ω-dodecanediol is a versatile material used as adhesive, crosslinking agent, pharmaceutical intermediates, and monomer for polyurethane, polyester. and it is high value-added product that can be biosynthesized from the inexpensive raw material, 1-dodecanol. Enzymes used in this case include P450 monooxygenases with ω-specific hydroxylation activity. In our previous study, α,ω-dodecanediol was synthesized at a conversion rate of 4.4% from 1-dodecanol. The aim of this study was to significantly increase the flux towards diol synthesis while reducing the production of the unwanted by-product lauric acid. As result, α,ω-dodecanediol was biosynthesized from 1-dodecanol at a conversion rate of over 70 %, and the production of lauric acid was significantly reduced. In addition, the applicability of wild and modified enzymes to several medium chain length primary alcohols was discussed based on the evaluation results of the ω-specific hydroxylation activity. |
