An Advanced Methodof Peptide Library Preparation for Ser/Thr Kinase Screening |

One-bead one-compound (OBOC) peptide library has been widely used for the screening peptide substrates of enzymes. For easy sequencing of OBOC peptide library by MALDI-TOF, ladder type OBOC peptide library has been developed, but isobaric amino acids such as Leu/Ile and Gln/Lys could not be distinguished by Mass. In this presentation, we prepared OBOC peptide library by using multiple capping method to distinguish these isobaric amino acids, and successfully screened peptide substrates for Ser/Thr kinase. |
