Fabrication of scaffold having a biocompatible and porous structure, mimicking the ECM, is important prospect in tissue engineering(TE) application. In this study, hyaluronic-acid(HA) and Pluronic(Plu) were chosen for the preparation of a hydrogel sponge. Although HA is a biocompatible material, it tends to easily degrade in culture medium or human body. To overcome this weakness, HA and Plu were acrylated and then photo-cross-linked by UV cross-linker. Finally, the cross-linked hydrogel is lyophilized and hydrogel sponge(HP-HS) is obtained. To determine potential of HP-HS as a support for TE, water up-take, degradation and biocompatibility studies were analyzed. From the results, FE-SEM image showed the porous structure of the hydrogel sponge which favors cell growth. KB cells were cultured in HP-HS and observed cell growth at during 7 days by MTT-assay and confocal microscopy. A substantial increase in the cell growth was observed after 7 days and were well adhered to the HP-HS |
