제목 Enhanced oral absorption of gold nanoparticle by using lactoferrin
초록 For extending application of gold nanoparticle (AuNP), its oral administration is very useful. But a significant limitation of AuNP is their low absorption efficiency in gastrointestinal tract. Therefore, we newly developed lactoferrin (Lf)-conjugated AuNP. In general, the lactoferrin is widely represented in various fluids, such as milk and saliva. Also, lactoferrin receptor (Lf-R) is overexpressed on the small intestinal epithelial cell for oral absorption. So, we adjusted AuNP modified with the Lf and PEG for improved half-life. When Lf-conjugated AuNP was orally administered into the mice, high accumulation of Lf-conjugated AuNP in bloodstream was observed. This Lf-conjugated AuNP did not show the cytotoxicity. Interestingly, because glioblastoma highly expressed Lf-R, the Lf-conjugated AuNP was targeted and accumulated to the glioblastoma in brain. As progressing experiments, we are doing the therapeutic effect of Lf-conjugated AuNP in the glioblastoma orthotopic animal model.
