Pancreatic cancer has been reported as a representative cancer with a poor prognosis and high resistance to chemotherapy. Considering that overexpression of endothelial growth factor receptor (EGFR), one of the hallmarks of pancreatic cancer, we devised a photo-immunotherapy using EGFR specific antibody-photosensitizer conjugate (EAPC). In EGFR-overexpressed cells, EAPC showed high cytotoxicity, resulting in increased level of immunogenic cell death (ICD) markers. In the condition that cancer cells and dendritic cells (DCs) were co-cultured, antigen-presenting markers were induced by EAPC, and thus upregulation of DC phagocytosis was confirmed. In vivo study showed that the number of DCs in lymph nodes was increased, and the migration of T cells and Natural killer (NK) cells in the tumor tissue was significantly upregulated. Therefore, these results suggested that EAPC could be an effective photo-immunotherapy for pancreatic cancer instead of conventional chemotherapy. |
