Phytosphingosine, a constituent of glycosphingolipids, is a common element of plasma membranes and mammalian tissues. We present here an efficient and stereoselective route for synthesis of D-ribo & D-arabino-phytosphingosine. The key step involves the OsO4-catalysed dihydroxylation of the N,N-diBoc and N-Boc protected allylic amino olefins derived from Garner’s aldehyde. Osmylation of the N-Boc protected olefin gave syn selective diol, whereas reversed anti selectivity was observed for N,N-diBoc protected olefin (ca. 20:1). |
