Process Development and Bioreactor Operation for the Production of hGM-CSF from Transgenic Nicotiana tabacum Suspension Cells |

In acidic conditions, the stability of hGM-CSF was deteriorated and protease activity in plant cell cultured media also showed relatively higher level. In bioreactor experiments, the optimum pH was determined at 5.5 for both the cell growth of Nicotiana tabacum and the production of hGM-CSF. Control of dissolved oxygen was solely achieved by aeration rate to investigate the effect on cell growth and hGM-CSF production and to avoid the effect of agitation rate. As a result, the cell growth was promoted by an increased DO up to 50% and any growth promotion did not observed above 50% DO. The production of hGM-CSF was enhanced by higher DO level, also. At 80% DO, 8.716 μg/L of extracellular hGM-CSF was obtained and which was a 2.5-fold increase compared to that of 20% DO. The higher DO was effective for the secretion of hGM-CSF. In addition, effect of aeration rate was examined. Although the cell growth at lag period was delayed at high aeration rate, the production of hGM-CSF could be significantly stimulated. With 0.3 vvm of aeration rate, 44.286 μg/L of hGM-CSF could be observed in the culture media. Compared to culture with 0.1 vvm, the value was 19.7-fold higher. |
