Transfection of Poly(4-vinylimidazole) in vitro and in vivo |
초록 |
Over the past decade there have been many attempts to design a nonviral vector to overcome the limitations of the viral vectors such as immunogenicity, oncogenicity, difficult production and purification, and target-cell specificities. In previous study, polymerization of 4-vinylimidazole(4V) prepared by decarboxylation of urocanic acid(UA) was carried out and used as a new gene carrier in vitro. It was found that the P4V formed a complex with DNA, and that the P4V/DNA complex exhibited very high transfection efficiency, through the proton sponge mechanism of the imidazole grows, and low cell toxicity. In this study, the transfection efficiency of the P4V/DNA complex was performed in vivo. |
