제목 A Novel DNA Carrier using Poly(4-vinyl imidazole)
초록 Poly(4-vinyl imidazole)(P4V) was used as the polymeric catalyst due to proton-donating property of the imidazole group in the side chain of the P4V. Also, side chain structure of the P4V is same as that of polyhistidine, which exhibited high transfection efficiency owing to pH-responsive endolysosome escape. It will be expected that the P4V is a new gene carrier with high transfection because the P4V has imidazole group in the side chain same as the polyhistidine. In this study, P4V was prepared as a new gene carrier based on high transfection efficiency and low toxicity on cells for possible clinical application. The P4V was synthesized by polymerization of 4V as new gene carrier. The 4V was prepared by decarboxylation of anhydrous urocanic acid. P4V was synthesized by polymerization of 4V with azobis(isobutyronitrile) (AIBN) as an initiator at 60℃ for 24 h under nitrogen. The prepared P4V was confirmed with NMR. The complex formation between P4V and DNA was confirmed by agarose gel electrophoresis. And the DNA in P4V/DNA complex was protected even at higher concentration of the nuclease than the physiological condition. The P4V showed less cytotoxicity than polyethylenimine (PEI). The P4V showed good transfection efficiency on several cell lines. As the results the P4V will be expected to use as gene carrier for gene therapy.
