In photodynamic therapy (PDT), photosensitizers produce singlet oxygen under the light irradiation and damage to tumor tissues. In many studies to increase the intracellular uptake of nanoparticles, fatty acids have been conjugated to the nanoparticles for enhancing uptake ability. In this work we investigated the effect of the alkyl chain length of fatty acids on the cellular uptake and cytotoxicity of nanoparticles. Thus, we used a poly(amido amine) dendrimer (PAMAM) as a carrier, which can help controlling the size of nanoparticles, and reduce toxity and immune reaction. The PDT agent, Chlorin e6 (Ce6), and fatty acids with different alkyl chain length were conjugated to PAMAM. Physicochemical characteristic of PAMAM-Ce6-fatty acid (PCFA)-conjugated nanoparticles was confirmed by 1H-NMR, FT-IR, UV-Vis, DLS, and TEM. Then, reactive oxygen species (ROS) generation and quenching effect were estimated. In vitro cytotoxicity and intracellular uptake were investigated. |
