A novel degradable branched poly(ester amine)s based on polycaprolactone and polyethylenimine as a gene carrier |

Gene therapy comprises a novel form of molecular medicine that will have major impact on human health. Polyethylenimine (PEI) is a nondegradable gene carrier with high transfection efficiency and high cytotoxicity. We synthesized low cytotoxic biodegradable poly(ester amine)s using low molecular weight PEI (0.6, 1.2 and 1.8 KDa) with a hydrophobic, biodegradable polycaprolactone diacrylate. The complexation of polymer/DNA was characterized by gel retardation assay, DLS and AFM to determine the complex forming ability, particle size and morphology, respectively. The copolymers were analyzed for their cytotoxicity and transfection efficiency on cultured HeLa, HepG2, and 293T. In complexation study, plasmid DNA was completely retarded at N/P ratio 5. In vitro transfection assay, using degradable poly(ester amine)s was quite comparable to that of PEI. Degradable branched poly(ester amine)s markedly showed low cytotoxicity compared with PEI 25K. |
