Synthesis of Poly(ethylene glycol)/ poly(trimethylene carbonate) AB2 Miktoarm Copolymers for Anticancer Drug Delivery |

Amphiphilic AB2 miktoarm copolymers that mimic structural characteristics of phospholipid show a great potential for drug delivery applications. In this study, a series of amphiphilic AB2 miktoarm copolymers consisting of hydrophilic poly(ethylene glycol) (PEG) as A block and hydrophobic poly(trimethylene carbonate) (PTMC) as B block were synthesized by ring-opening polymerization of TMC using methoxy PEG as a macroinitiator. The chemical compositions of PEG-PTMC2 miktoarm copolymers were confirmed by 1H-NMR, GPC, and FT-IR measurements. The miktoarm copolymers, PEG2k-PTMC2k and PEG2k-PTMC4k, readily formed spherical micelles (40~50 nm) when they self-assembled in aqueous solution. Paclitaxel (PTX) was efficiently loaded into PEG-PTMC2 micelles. The drug loading capacity and release characteristics of the miktoarm copolymer micelles were evaluated in comparison with comparable diblock copolymer micelles. |
