제목 Amphiphilic copolymer nanoparticles for targeted DNA delivery
초록 Genes are very attractive candidates for therapeutic use in a variety of disease states due to their ability to produce bioactive proteins using the biosynthetic machinery provided by host cells. Self-assembling polycation/DNA complexes represent a promising synthetic vector for gene delivery. The amphiphilic copolymer composed of hydrophilic poly(ethylene glycol)(PEG) and hydrophobic ε-caprolactone(CL) was synthesized to form nanoparticles. For the ionic conjugation between DNA and the copolymer, poly(lysine) with amino groups was grafted at the end chain of the amphiphilic copolymer. For the evaluation of the polymer synthesis, we examined GPC, 2,4,6-trinitrobenzene sulfonic acid (TNBS) assay. We determined nanoparticles/gene complex ratio, cytotoxicity, transfection efficiency for the evaluation of the DNA delivery.
