Loop-mediated isothermal amplification (LAMP) is one of the isothermal gene amplification methods that has great advantage, because it is more suitable for point-of-care testing and can be performed in simple, miniaturized settings. Herein, we designed a simple and rapid assay based on polydopamine nanoparticle (PDAN) as quencher. Fluorescein-modified probe interact with the PDAN through π–π stacking and is quenched by FRET mechanism. Fluorescein-probe could be recovered the fluorescence after hybridization with nucleic acid target amplified by LAMP, since the interaction between double-stranded DNA with PDAN was weakened due to the changed conformation. We performed the LAMP reaction for the antibiotic resistance gene KPC, followed by addition with fluorescein-probes specific to the loop region of the amplified DNA products that are adsorbed on the surface of PDAN. The recovery of fluorescence intensity was characterized by visualization as well as spectral quantitative analyses. |
