Delivery of microRNA using polypeptide-based cationic polymer for ischemic stroke |

Polymeric non-viral carriers have advantages such as convenience of handling and flexible gene loading capacity over viral vectors. Despite these advantages, their applications to human gene therapy are still limited because of their cytotoxicity and low transfection efficiency. To overcome these limitations, novel cationic and biodegradable polymer (Methoxypoly(ethylene glycol)-b-poly{N-[N-(2-aminoethyl)- 2-aminoethyl]-L-glutamate} (mPEG-b-PNLG) was synthesized for gene delivery. A series of mPEG-b-PNLG polymers having different molecular weights of PEG and side chain lengths of PNLG were used to deliver microRNA by forming nanocomplexes through electrostatic interactions. These nanocomplexes enhanced the cellular uptake of microRNA, resulting in greater transfection efficiency compared with bPEI/microRNA complex. So, these novel cationic polymers can be used as effective non-viral polymeric carrier for microRNA delivery. |
